3. Product Description
The Kit uses sterile buffer in Transparent Eppendorf tube and indicator dye in Amber colored Eppendorf tube. In addition, a sterilized cotton swab was given for taking the samples from the surfaces.
4. Storage and Handling Requirements
Store all reagents at below 25°C (both un-opened and in-use product) and use the reagents within 3 months once opened.
Reagents should not be used past their expiration date.
Shake the reagents briefly before each use.
5. Warnings and Precautions
For any food contact surfaces use only.
One tube for single use only.

6. Principle
The principle of this assay is that the binding of protein molecules to indicator dye under acidic conditions results in a color change from yellow to green.
7. Test Methods
At first, dip the cotton swab into the buffer solution (Transparent Eppendorf tube), then wipe the detection area (100 cm2) by that cotton swab, and finally dip that swab into the detection solution (Amber colored Eppendorf tube). ()
8. Limit of Detection (LOD)
The LOD study established the lowest concentration of protein molecule (40 µg/100cm2) that consistently yielded a 95.65% positivity rate with the hygiene detection kit compared to standard culture method.
9. Results interpretation
If the cotton swab turns from yellow to green color means the surface is contaminated with microbes. If the cotton swab shows black-ash color means the surface is contaminated with dust. If the cotton swab shows same color (yellow) means the surface is clean adequately. Now, depending on the intensity of green color the cleanliness varies. The more the green color appears the more unhygienic the surface is, thus extensive cleaning the surfaces is required.

সংরক্ষন এবং হ্যান্ডলিং প্রয়োজনীয়তা
সমস্ত বিকারক ২৫ ডিগ্রি সেলসিয়াসের নীচে সংরক্ষণ করুন (খোলা এবং ব্যবহারযোগ্য উভয়পণ্য) এবং একবার খোলার ৩ মাসের মধ্যে বিকারকগুলি ব্যবহার করা উচিত।
বিকারকের মেয়াদ শেষ হওয়ার তারিখের পরে ব্যবহার করা উচিত নয়।
প্রতিটি ব্যবহারের আগে বিকারক অল্প ঝাঁকিয়ে নিতে হবে।

এই পরীক্ষার মূলনীতি হল যে অম্লীয় অবস্থার অধীনে সূচক রঞ্জকের সাথে প্রোটিন জাতীয় কণার বিক্রিয়ার ফলে হলুদ থেকে সবুজ রঙে পরিবর্তন হয়।
পরীক্ষণ পদ্ধতি
প্রথমে তুলার দণ্ডটিকে বাফার দ্রবণে (স্বচ্ছ এপেনডর্ফ টিউব) ডুবিয়ে দিন, তারপর সেই তুলার দণ্ড দ্বারা সনাক্তকরণ এলাকাটি ((১০০ সেমি২) মুছুন এবং অবশেষে সেই তুলার দণ্ডটিকে সনাক্তকরণ দ্রবণে (রঙিন এপেনডর্ফ টিউব) ডুবিয়ে দিন । (চিত্র)
সনাক্তকরণের সীমা
সনাক্তকরণের সীমা গবেষণায় প্রোটিন জাতীয় কণার সর্বনিম্ন ঘনত্ব (৪০ মাইক্রোগ্রাম/১০০সেমি২) প্রতিষ্ঠিত হয়েছে যা মানসম্মত স্ট্যান্ডার্ড পদ্ধতির তুলনায় হাইজিন সনাক্তকরণ কিটগুলির সাথে ধারাবাহিকভাবে ৯৫.৬৫% ইতিবাচকতা হার দিয়েছে।
ফলাফল ও ব্যাখ্যা
যদি তুলার দণ্ডটি হলুদ থেকে সবুজ রঙে পরিণত হয় তার মানে হল পৃষ্ঠটি জীবাণু দ্বারা দূষিত। যদি তুলার দণ্ডটি একই হলুদ রঙ দেখায় তবে এর অর্থ পৃষ্ঠটি পরিষ্কার। যদি তুলার দণ্ডটি কালো-ছাই রঙ দেখায় তবে এর অর্থ পৃষ্ঠটি ধুলো দ্বারা দূষিত। এখানে পরিচ্ছন্নতার পরিমাপ নির্ভর করে সবুজ রঙের তীব্রতার উপর। যত বেশি গাড় সবুজ রঙ প্রদর্শিত হবে পৃষ্ঠটি তত বেশি অস্বাস্থ্যকর বুঝতে হবে, তখন পৃষ্ঠগুলি ভালোভাবে পরিষ্কার করা প্রয়োজন।
